If you have enough medium in each well you can fairly easily harvest off 
the top without disturbing the cells on the bottom.  I fill up my wells 
  all the way and then can harvest up to 250uL from each (using a 
multiwell piptettor and collecting into a fresh 96 well plate).  Then 
refeed with fresh medium.
