I would like to know if anyone has a good protocol for whole
cell and/or nuclear crude protein extracts from Chlamy that would be
suitable for use in footprinting or band shift experiments with
NUCLEAR genes. I have tried adapting a whole cell extract procedure
from soybean literature, but I'm not very enthusiastic about the
results so far. Since isolating nuclei is rather straightforward, I
think nuclear extracts will be more sucessful, but I will still need
to look for some proteins that are likely in the cytoplasm, not the
nucleus.
