I am attempting to isolate a nuclear protein from Chlamydomonas
reinhardtii. The nuclear prep in the Chlamydomonas Sourcebook works
great (except that my pellet is still green). However, when I try to
isolate the protein, I end up with a pellet so sticky that I cannot
solubilize it. I am incubating it in 0.02N NaOH and then in 30% TCA.
Before the TCA the pellet is impossible to get into solution. Does
anyone have any ideas as to how I could isolate a nuclear protein from
chlamy? I would appreciate any help! Thank you in advance.
