I'm going to isolate gamete autolysin.
According to the Chlamydomonas Sourcebook (page 593), wild-type cells of
both mating types should be pregrown in liquid phototrophic cultures, and
then transferred to nitrogen-free medium.
Is the choice of growth medium important? Phototrophic or mixotrophic
growth (i.e. without or with acetate)? High-salt minimal medium, HSA or TAP?
Is "nitrogen-free medium" HS, HSA or TAP with no ammonium chloride?
