I am finding low levels of motility in logarithmically growing chlamy
cells. Motility levels are about 10-50% in mid-log cultures. I am
using the UTEX 2244 strain, cultured in Sager-Granick with the
Hutner's trace metals. Cells are grown at 25C & 7500 lux. Mid-log
densities in this medium are around 5e5/mL to 1e6/mL. Is this
common? Non-motile cells in these cultures are generally in doublets
or quadruplets. They appear similar to the palmeloid state discussed
in the chlamy handbook. Any advice on getting higher motility - e.g.
90% or more of cell swimming?
