Have you tried the protocol in:
Gloor, G. B. and W. R. Engels, 1992 Single fly DNA preps for
PCR. Dros. Inform. Ser. 71: 148-149.
The article is also in the first issue of DIN, available on FlyBase, and it
is described as well in (Gloor et al., 1993, Genetics 135:81-95). WeÕve
used this method on close to 10,000 individual flies now. The failure rate
is probably less than 1% for us.
