Southern blot hybridization

Help !!! I am having all kinds of problem with southern blot 
hybridization. I am new on Drosophila, and I would like to know some 
basic things as how much DNA in general one load in a slot for 
hybridizing with a single copy gene. The specific activity of the probes 
used and so on...please, anybody with experience in hybridization tellme 
about your experience, because I am driving crazy with frustrating 
results on such basic step !!!
