The consensus appears to be: 1) After harvesting and popping vitelline membranes, destroy endogenous HRP with a H2O2/MeOH step (3% 15 mins). 2) Use fresh DAB made from powder rather than the premade Vector stuff. This can lower background. 3) Ni2+ can increase background. Avoid if possible. Martin
