Study of a region

Hi,
I have two questions: first, does anybody know if Drosophila
transformation with "cosmid vectors" (large inserts) have ever worked
well ? Do you have references on this subject ?
Second: we are analyzing genomic sequences in order to determine the
number of genes in a given region. Do you have an idea of which
softwares are the most appropriate in this respect ?
