Hi--
I've been trying to isolate sequences flanking a PlacW element, which I
believe resides in the X chromosome centromeric heterochromatin. I've
used the
"inverse PCR protocol" (digesting genomic DNA, ligating and using inverse
PCR primers), with no success (positive control works!). I am wondering
whether this is due to the centromeric heterochromatin that flanks the
pelement. Has anyone successfuly rescued a pelement from heterochromatin,
if yes, which method was used?
