Hi Hong--
The Bloomington Stock Center maintains a duplication with the same
breakpoints as Dp(1;Y)g[+]. It is Dp(1;f)LJ9, in stock
3219 C(1;Y)1, Df(1)g, y[1] f[1] B[1]/C(1)A, y[1]/Dp(1;f)LJ9, y[+] g[+]
na[+] Ste[+],
which duplicates 12A6-10;13A2-5 and the tip of the X (1A1;1B3-4). The
12A;13A region is subdivided by Dp(1;f)LJ8, in stock
2870 C(1;Y)1, y[1] g[2] na[1]/FM3/Dp(1;f)LJ8, y[+] g[+] na[+] Ste[+],
which duplicates 12E1;13A2-5 (and perhaps a bit more distally) and the same
X tip region.
These two duplications were derived from In(1)sc[29] (1B3-4;13A2-5) by
deleting everything between the distal bands and the centric
heterochromatin. Dp(1;Y)g[+] and Dp(1;Y)na[+] were probably made the same
way, except that In(1)sc[29] was first crossed onto the attached XY
chromosome C(1;Y)6 (also known as "XYL.YS"). Irradiating a In(1)sc[29] +
C(1;Y)6 chromosome would be an efficient way to make a series of Dp(1;Y)
chromosomes if you need to duplication map your mutation more closely. You
can read a really nice description of a screen like this in a DIS article
from Juan Botas and Brenda Lilly ("An efficient mutagenesis screen to
generate duplications of polytene section 8 on the X chromosome of
Drosophila melanogaster", D. I. S. 1999 82:105--112). You can also look at
http://flybase.bio.indiana.edu/.bin/fbpcq.html?FBrf0125111 for a brief
description of a screen we did.
