(*= cross)
If the P-element is on X chromosome

Cross I Cross IA
P/P * FM3/FM7 FM3/FM7 * (transposase) delta 2-3 kinked sb
(red eye color)

Cross II
P/FM7 * +/FM7 ; +/625
(patchy red eye color)

Cross III
    			P/FM7 *FM7male
			(orange eye)

Cross IV
P male * FM3/FM7 (as many crosses as possible)
           		(white eye flies)

Cross V P/FM7 * P male (self crosses from each vial)
			(white eye flies)

The homozygous lethal lines or homozygous viable with phenotype will be the p-element excision mutants

(*= cross)
If the P-element is on II chromosome

Cross I Cross IA
P/P * cyo/ tft cyo/tft * (transposase) delta 2-3 kinked sb
(red eye color)

Cross II
P/cyo * +/cyo ; +/625
(patchy red eye color)

Cross III
    			P/cyo * cyo/tft
			(orange eye)

Cross IV
P/cyo * cyo/tft (as many crosses as possible)

Cross V P/cyo * P male (self crosses)
			(white eye flies)

The homozygous lethal lines or homozygous viable with phenotype will be the p-element excision mutants

(*= cross)
If the P-element is on III chromosome

Cross I Cross IA
P/P * TM3sb/TM6B TM3sb/TM6B * (transposase) delta 2-3 kinked sb
(red eye color)

Cross II
P/Tb * 625/Tb
(patchy red eye color)

Cross III
    				P/Tb * TM3sb/TM6B
				(orange eye)

Cross IV
P/Tb * TM3sb/TM6B (as many crosses as possible)

Cross V P/Tb * P male (self crosses)
			(white eye flies)
The homozygous lethal lines or viable with phenotype will be the p-element excision mutants

Hi Drosophilists,

Could any of you guide me (schematic) to generate imprecise or precise excisions or deletions using p-element insertion lines available in the stock centres?

Any help would be highly appreciated.

Sincerely, Venkat
