GFP and nail polish

Steffen,

I've tried a number of methods for live cells (nail polish, agar, pva)
and none really helped.  In the end I got an 'Autofluor Cell Chamber'
from Molecular Probes, mounted the coverslip in that, filled it up with
medium and put a 3.5cm petri disk lid on top (excluding any air
bubbles).

The whole assembly is them inverted (if using an upright microscope
rather than inverted) and mounted on the Confocal Mic stage.

For fixed cells I just mounted in Moviol.
