I am currently trying to make a primary culture of keratinocytes (or
epidermal cells, generally) from murine ear and/or dorsal skin.
I have been using RPMI + 5% foetal calf serum + gentamicin (100U/ml) +
2-mercaptoethanol (0.1%) + glutamine (2mM) as a culture medium, but am
finding that cell growth is very slow, and not satisfactory for the
purpose of my experiments.
Is there anyone who has successfully cultured keratinocytes, who would be
able to give me some advice regarding "magic ingredients" for culture
medium, or specific techniques that have been successful?
