Help : in Vitro Ig V gene production

We are trying to use the new vector pZeoSV (inVitrogen) to express
stable in Vitro Ig V genes. We have come up against two problems:

1 - low concentration of protein (~10 ng/ml in culture supernatants)

2 - sensitivity to Zeocin varies from one culture to another (using
250 µg/ml Zeocin in RPMI medium + FCS 10%)

Suggestions would be welcome.
