OK, here's the scoop. I am trying to do a T cell proliferation assay
where I immunize mice with a protein, remove the draining lymph
nodes, and culture the T cells with antigen. Proliferation is
measured by 3H thymidine incorporation. If I use the protein as
the in vitro antigen, it works fine. If I use a synthetic peptide
of the protein (one that works for other labs and for a prior grad
student in my lab) it doesn't work. I'm really beating my head on
the wall with this and I'd appreciate any suggestions at all.
