NEED HELP WITH T-CELL HYBRIDOMAS

I am about to make antigen specific T cell hybridomas after imunization with a 
peptide.

What is the frequency of Ag specific hybridomas I can expect if I start with 
the draining lymph node and a 3-day restimulation in vitro with the peptide ?
Is there a way (facs sorting ?) to select for T cells that have responded in 
vitro before doing the fusion? Would that help anyway ?
