Any particular reason to use popliteal lymph nodes? I assume you are
using naive mice, becuase when I immunize in the footpad with Freund's
I get huge nodes with 1-2x10^7 cells per animal. If you need cells from
an unimmunized animal can you try the mesenteric lymph nodes? Bigger
and easier to get out; more cells.
