I am a developmental biology student doing
whole-mount in situs with an antibody that is
sensitive to most types of fixation methods.
At the moment it seems to be happy with a methanol/
DMSO fix, however this means that my embryos are
undergoing massive shrinking during the protocol, and
we are concerned that the unfolding and refolding
of all the proteins around is resulting in a matrix
that is partially impenetrable to the antibody.
Any ideas would be greatly appreciated.
