purification/Western blotting/identification of mucins

Can anyone give me some advice re the following:-

1) Is there a good/efficient way to desalt and concentrate mucin containing
solutions?

2) Is there a trick to get good separations of mucins on SDS PAGE?

3) Is it neccessary to use any particular membrane/blocking
solution/protocol/ etc when Western blotting?

4) How easy is it to get N-terminal amino acid sequence from heavily
O-linked mucins? - should you deglycosylate first?
