Amplification of monoclonal by passage through mice

We have been using a monoclonal antibody which we harvest from
the hybridoma cells. However, for some purposes, the titer is too
low. We have recently tried injecting hybridoma cells into isogenic
mice (after pristane priming) to produce ascites according to the method
in Harlow and Lane but this hasn't worked. Does anyone have experience
with this procedure and suggestions for getting a response in the mice?

Or, does anyone know of a better method of concentrating our antibody?
We have tried the EZSep kit made by Pharmacia and found that it did not
concentrate the antibody at all (although it did remove BSA pretty well).
If anyone has any suggestions for us, we would be delighted to hear from
you.

Thanks in advance,
