I am trying to develop an immunochromatographic assay to a small molecular
weight chemical, about 200-400 M.W. The reporter is a monoclonal
conjugated to colloidal gold.   So far, I've been having problems with the
reagent binding to the membrane, even though it's coupled to BSA.  I've
tried millipore nitrocellulose with variable results, and also S&S with
better results.  However, so far I've been unable to detect the chemical
at low ng/ml levels.  Technical help from manufacturers suggest trying
various membranes and pore sizes.

Has anyone else ever tried to develop an assay such as this?  Can you
offer any insight into how to approach this.
