mRNA for IFN in 3T3 monolayers

We are trying to induce IFN alpha production in mouse 3T3 cell monlayers
using poly I:C.  Our goal is measuring mRNA, not IFN in the supernatant.
If anyone has used this or a similar technique, we would appreciate any
advice you might have.  Issues we are curious about are duration of Poly
I:C, addition of actinomycin D or not, culture time, and harvest
conditions.  Thanks in advance...
