Phenotyping mouse brain cells in situ

We are immunophenotyping normal mouse brain cells by
flow and direct staining of frozen and fixed brain
tissue. We can easily detect certain phenotypes by
flow but are unable to detect such cells in fixed or
frozen sections. One possibility is that cells in
the sections are 'coated' with substances that are
removed during preparation of the cell suspensions.
Any one have a similar experience? Any solution to
in situ detection? All suggestions appreciated.
