Since the MTT assay is dependent on the oxidative capacity of the cell it
tends to relate very well to the mitochondrial content of the cell.  As a
result I have found that it works well with cells in culture or actively
proliferating primary cells such as T cell blasts.  Where I have found that
it works poorly is on primary cells like thymocytes and primary fibroblasts
or on the first few rounds of T cell replication after mitogen stimulation. 
You can still get a statistically significant window with these cells if you
wait long enough.
