Staining tissue for Macrophages

I have suspected xanthoma lesions (bird) which should contain macrophages
and multi-nucleated giant cells if a TRUE xanthoma. Suggestions for
staining 6 micron thick sections (cryostat).

1.Should I fix the sections first? What to use?

2.Is the cold temperature storage destroyed by WBC's etc. (i.e. -70 C)

3.What staining to use on the tissue? Tried Wrights, Hematoxyolin, Giesma
etc. with no luck.

Thanks. SG
