Pharmacia the giant Swedish company that produces sepharose, sephedex,
sephacryl and all, also sells a series of booklets (about $15 each)
explaining the principles of various Pharmacia products. I remember
one booklet on Sephadex and molecular sieving, another on ion exchange
chromatography and so on. They give you practical hints on how their
various products are used as well. I don't have any experience using
affinity purification columns so I can't be more specific. If your
company orders directly from Pharmacia, getting these booklets are
simple. Or if you purchase their products indirectly from another
distributor - say perhaps Sigma Chemical which everyone orders from
one time or another, check with Sigma directly about availability.
