NK cell Isolation

Hello,

I am a 3rd year undergraduate at Dalhousie University taking an advanced
immunology course. This course, called "the Granulocytes and the Immune
response" requires me to write a research grant - one that could be
submitted to a granting agency. Sadly, I have no background
in experimental immunology.

My grant involves the isolation of NK cells. The specific protocol that I
am using avoids the use of plastic adherence to remove monocytes since
that involves incubation at 37 C. The authors of a particular paper
(Taub et al. 1995. alpha and beta chemokines induce NK cell migration
and Enhance NK-mediated cytolysis. J. Immuno. 155:3877-3888) claim that
such incubations "appear to inhibit chemokine-specific NK cell
responses...by down-regulation of chemokine receptor expression on
lymphocytes." Have other investigators noticed this effect since it is
the only paper that I have found that mentions this.

Also, is there a good way to label NK cells (i.e.: do they take up 51Cr?)
for a chemotaxis essay?

Answers to these questions can be sent to me at the following email address:

Thank you for the time.
