Ask for gene knockout protocol

Our Lab. just cloned a novel molecule which function is still not very clear. So 
using the gene-knockout model is one of the useful methods to identify its 
functions. I am very greatly appreciated if anyone could provide me with the 
gene-knockout protocol, any good suggestions, or you could provide me with the 
e-mail address of expert in this area.
