I wonder if somebody else has observed a reduction of phycoerythrine
fluorescence upon paraformaldehyde fixation. In an attempt to measure
antibody clusters in heparinised blood by flow cytometry, I used
a distilled water lyse as commercial systems did not perform
satisfactory. Unfortunately all PE markers tested so far (cd14, cd45ro,
cd 4) show significantly higher fluorescence compared to cells treated
with commercial lysing systems which leaves me with problems regarding
compensation. So I will try to compare expression levels in gradient
blood and in unlysed samples over the next few days, I thought somebody
might have another idea.
