I have made T cell clones from frozen PBMC in the past. The main problem is
that you run out of autologous antigen presenting cells to use rather
quickly. You can spin things out a bit longer by doing some stimulations
with allogeneic cells and PHA. See International Immunology vol. 8, p.1877
for our protocols.
As with all T cell cloning, the rule is make 'em and use 'em before the
little buggers die on you.
