Megan, since this is a mouse-model, the first approach is to work with 
fresh-frozen tissues.  these will be positive if your antibodies are good, as 
you say there are many antigen retrieval protocols and it best 
approached empirically.  If no protocol was done on your specific tissue, 
than trail and error is the way to go,  good luck
