Theresa,
Production of monoclonal Abs is definately more efficient by ascites
than by tissue culture. Hybridomas in tissue culture generally yield
about 50ug/ml of specific antibody, while ascites yields are typically
1-10 mg/ml. Problems with ascites production include variability from
mouse to mouse in total Ab yields, and the need for mice of specific
ages. Of course, you also need certified animal rooms. This makes
ascites production an expensive undertaking. However, if the animal
facilities are already in place, ascites will generally give you more
antibody than tissue culture.
