For lymphocyte isolation try the classical method of density gradient
centrifugation with a Ficoll (or Percoll) step of 1.077 g/ml.
For hints regarding your double-staining I should know more about your
antibodies, e.g. species and isotypes of primary and secondary
antibodies.
