Look up Paul Lacy papers from the 80's.  They developed a protocol for 
isolating islets by perfusing pancrease-in situ with enzymes.
good luck its not easy, but maybe the techniques have been improved upon. 
 Do you really need islets could you use transformed cells for your 
studies?
