I am not sure what to do with my immunization. I have a recombinant
protein, containing a 6xhis-tag at the N-terminus. The protein is
isolated and purified under denaturing conditions, but I have problems
removing the his-tag.
Does anybody have experience in immunizing animals with a protein
containing such a tag ? I mean, may such a tag be responsible for
a low antibody production against the protein of interest ? Is it
possible to guess the antigenity of a 6xhis-tag compared to a protein
of 170 amino acids ?
