lysozyme plate assay

Hello,

I am planning to assay chinook salmon plasma for lysozyme concentration
using the agarose plate technique.  Would anyone be able to clarify for me
the following:

1.  What pH should the sodium phosphate buffer be? (I have seen in the
literature a range of 5.8 to 6.24)

2.  What is the wavelength at which the turbimetric assay (to determine
lysozyme activity) should be performed (I have seen both 530 and 450
referenced)
