I am begining the studies of T cell responses to a self antigen which elicits an antibody (IgG) response in individuals who lack this self-protein due to genetic defect. Healthy individuals are also known to make antibodies to this self-antigen.
I set up LTTs with and without depleting CD8+ cells from PBMCs of healthy individuals. I was expecting better thymidine uptake in cultures with depleted CD8+ cells but the results were contrary, i e better respose in undepleted PBMCs. I got similar results in three different blood samples, I have yet to see the patient responses.
I would welcome possible explanations.
