3h-Thymidin versus BrdU- Proliferation-assay

I have found that the BrdU ELISA is not as sensitive as Thymidine, but it
does have its place, most notably when BrdU is used on the FACS. You can do
a whole blood proliferation assay by flow cytometry; this is especially when
you are not sure exactly what the antigen is, for example, using cell
lysates. Whole blood can be used as the source of effectors and antigen
presenting cells (APC) and so just by adding BrdU and some clever staining
you can follow proliferation of any cell type you are interested in.
Thymidine just can't do this, but is more sensitive. Consequently, if you
know the antigen and have a source of APC this is the method of choice. So
the answer is choose the one which is most appropriate to the assay you are
trying to conduct!
