[Immunology] Differentiation of macrophages from PBMC

Hi all,
Can Somebody suggest me a method to generate macrophage from PBMC. 
1) Often the protocols describe a simple method of plating monocytes
(seprated from PBMC by adhesion to plastic) and culturing them in RPMI
for 7 days. I have used THP1 cell line which upon addition of PMA gets
converted to Macrophage. I wonder wthether PMA should be added to
monocytes also? How does PMA work? 
2) How can I then confirm the differentiation of monocytes to
macrophages?
