I am having problem with my peripheral blood mononuclear cells
(PBMCs) separation. I used LymphoprepTM (Axis-Shield), which the method is
basically based on gradient density separation. The yield was unsteady, 2-3
times I have good yield, where the white cloudy layer (The PBMCs) was
distinctly separated. Using the same gradient medium, sometime does not
yield anything, I observed no white cloudy layer at all.
It should be a simple protocol, I think I might have get something wrong
with it. Troubleshooting and guessing the possible reasons.
