The project, described in the attached summary, will construct normalized libraries from several pools of RNA, end sequence 40,000+ clones from each species, re-array and fully sequence ~ 12,000 representative FL clones from each species. Both the sequences and clones will be freely available, the sequences through the TIGR web site and GenBank, the clones from the Noble Foundation in the USA and from INRA-CNRGV, Toulouse in Europe. While preparing the proposal, I had already solicited offers of RNA donations from a number of you. Now that we are ready to get started, I would like to ask any of you who have some RNA to spare if you would consider providing some to go into one of the library pools. Ideally we would like 5-10 ug of total RNA, but could get by with as little as 1-2 ug. We are looking for RNA in the following categories, where the numbers indicate the intended pool: Flowers 1 Early seed 1 Late seed 1 Stems 1 Leaves 2 Biotic stressed leaves 2 Abiotic stressed leaves 2 Tissue culture 2 Elicitor-treated plants 2 Roots, early nodules 3 Roots, late nodules 3 Roots, biotic stress 3 Roots, abiotic stress 3 Within the biotic and abiotic stresses, we would be happy to have RNA from you favorite stress(es) - the more the merrier as we try to maximize the diversity of the libraries. Since one of the goals of the project is to support genome annotation, there is a strong preference for RNA samples from the varieties being sequenced (A18 for Medicago, MG-20 for Lotus and Williams for soybean), but RNA from other accessions may also be useful. In order to get this project rolling, please send us some of your samples ASAP. Shipping at room temperature under ethanol should be fine. Dry ice if you prefer. We can provide a FedEx/DHL account number if you need it. If you are shipping from abroad, please contact me first so that I can send you a Toxic Substances Control Act (TSCA) certification to include with your package and speed it through customs.
