[Medicago] chloroplast-free DNA

Hi

 

I'm trying to isolate a lot of CP-free DNA for sequencing without going
to the trouble of nuclear isolation.

I've germinated seed in the dark and collected hypocotyls (about 6g).
However, based upon a similar approach with Brassica, I don't think I'll
get enough DNA. I suspect that compared with leaves, the hypocotyls have
big cells and thus fewer nuclei per gram

 

Any suggestions to get increased CP-free biomass?
