Hi Brian-

#1  Take a look at Marchuk et al (1991) NAR 19: 1154.  This is an easy way to 
make a T-vector.  No restriction needed.  This works pretty well for us.

#2  I'd recommend putting 5'GC 3' in front of the restriction site.  This is
the most thermally stable dinucleotide.  This has worked well for us with
EcoR1 and BamH1.
