We have recently found that after cloning PCR products into the SmaI site of bluescript some or most of the linker sites have been lost. has anyone else had this experience and is there a reason? We do blunt end our PCR products before subcloning. Kathy Cheah HRMBDKC at HKUCC.BITNET
