filamentous phage RF

Hi netters,


I have a problem with the preparation of replicative
form of fuse5 (a derivative of the filamentous phage fd-tet. What happens 
is that after the bacteria have grown under selection pressure, 
(meaning the phage must be there) only minute amounts of DNA are 
recovered from 0.5 l of culture, just enough to get 
ca. 100 colonies after electroporation. The protocol that is used is
exactly what George Smith writes in his manuscript. With other, similar
phages, this problem does not occur.

Does anybody have a similar problem or a suggestion?
