Use less template DNA (25 ng should be enough for all but the largest hyb volumes) and take good care of your Klenow! If 5 people in a lab each put the enzyme
on ice for 15 minutes ("just a little while...") while they plan their experiments, it doesn't take long for the enzyme to die. Better still, make up your own
kit; it won't necessarily work better, but will force you to really learn about
what's going on.
