I have been trying to use the Amersham ECL (RPN 3000 and 3004) to directly screen
colonies. After having carried out several tests, we found out that peroxydase
hybridizes unspecifically to the DNA of clones creating a high background which
enables us to detect specific hybridization signals.
Has anyone got any experience in this type of screening and how could I decrease
this unspecific signal of peroxydase ?
