lon mutation - hints on harvesting slime?

We have incorporated the lon protease mutation into an E. coli host
strain that we are creating. We hope to express some eucaryotic enzymes
in this background.

The lon mutation makes strains slimey.

Does this cause any problems in harvesting soluble protein?

Our standard way of harvesting soluble protein is to sonicate, either
with or without a lysozyme treatment. Does anyone have suggestions
for improving this?

Thank you. Russell Malmberg [russell at dogwood.botany.uga.edu]
