PCR Mutagenesis

I couldn't find but one other reference to the use of PCR in random mutagensis,
Journal of Biological Chemistry 265 (33)p20085. It just shows that 25 cycles
on a 480bp fragment generated phenotypic mutants at around 4%. This of course
does not indicated changes at the DNA level.


If anyone has access to the now famous journal "Technique", could you please
post the Mn+ levels used to decrease Taq fidelity ? If possible the reaction
conditions used and the resulting misincorporation statistics ?
